"Hard head" disease is a common physiological disease in the fruit of the golden pear variety, but little is known about the mechanisms involved in its development.

In November 2017, Associate Professor Yang Shaolan from Qingdao Agricultural University published an article on Cell Calcium entitled “The changes of intracellular calcium concentration and distribution in the hard end pear (Pyrus pyrifolia cv. 'Whangkeumbae') fruit”. The relationship between the hard-end dysregulation and calcium deficiency in pear fruit.

The Ca 2+ flow rate, Ca 2+ distribution, and Ca, Mg, K, B, and N contents of pear fruits were determined. The results showed that during the fruit development, the contents of Ca/N, Ca/K, Ca/Mg and B in the "hard head" disease pears were significantly lower than those in the normal group. However, no calcium deficiency was detected in the soil and leaves in the environment in which the "hard head" diseased pears grew.

In addition to detecting the element content, the NMT biophysiology detector Physiolyzer® based on Non-invasive Micro-test Technology (NMT) was used to detect the normal fruit and different parts of the "hard head" diseased pear fruit. Ca 2+ flow rate. The data showed that the Ca 2+ uptake of the “hard head” disease pear flower bud was significantly lower than that of the normal group after flowering for 90 days. The content of free Ca 2+ and Ca 2+ in the fruit cells of the “hard head” disease pear was lower than that in the fruit development stage. Under normal conditions, the opposite trend appeared after 120 days of flowering.

Detection of Ca 2+ flow rate in different parts of different pear fruits. Negative value means absorption

The study not only from the perspective of Ca 2+ flow, but also combined with experiments on gene function verification, the results show that the decrease of Ca 2+ absorption leads to the decrease of Ca 2+ into the “hard head” disease pear, leading to the imbalance of intracellular Ca 2+ , thus causing The occurrence of hard end disorders.
