As a branch of in vitro diagnosis, liquid biopsy can monitor circulating tumor cells (CTC), circulating tumor DNA (ctDNA) fragments and exosomes (Exosome) released into the blood by tumors or metastases with its non-invasive blood detection technology. It is a breakthrough technology for detecting tumors and adjuvant therapy. In the early days, people paid insufficient attention to liquid biopsy and did not find its potential application value. Therefore, the field progressed slowly in the decades after the concept of circulating tumor cells, but with the deepening of its research, people gradually realized that The huge application value, the research on liquid biopsy at this stage shows an explosive growth trend (Figure 1), and the major biotech companies have shown great interest in the field, and their development efforts have also increased. It has gradually entered the clinical stage.

Liquid biopsy techniques mainly include CTC, ctDNA and Exosome. CTC is a tumor cell released from solid tumors or metastases and enters the peripheral blood circulation. It focuses on cytopathological diagnosis and is suitable for monitoring the effect of chemotherapy. ctDNA is a DNA fragment released into the blood circulation system by tumor cells, focusing on the tumor gene level. Mainly reflected in the gene mutation information of tumors, more suitable for targeted drug therapy; and Exosome sources, lymphocytes, dendritic cells, mast cells and epithelial cells, tumor cells, etc. can secrete exosomes, including The protein contains over-expressed specific protein markers, and also contains a large number of miRNAs, cirRNAs and DNA fragments. The biological information contained is very rich, but the current research is not deep enough, and its clinical application remains to be discovered. Compared with traditional methods of tumor diagnosis, liquid biopsy has the advantage of solving the pain point of precision medical treatment , reducing the risk of biopsy through non-invasive sampling, and can effectively prolong the survival of patients, and has a high cost performance.
Since the advent of liquid biopsy, various new technologies have been developed around it to promote its early clinical application. Especially for CTC enrichment and analysis techniques, membrane filtration and immunomagnetic beads have been positive. Negative enrichment, microfluidics, immunofluorescence staining, RT-PCR, etc., the continuous innovation of these technologies has improved the accuracy of obtaining CTC from massive blood cells and provided support for subsequent analysis, and also made the clinical clinical Application is possible. But we should also see that although there are all kinds of new technologies emerging, each of them has its limitations. We have summarized them (Figure 2).
