Recently, the internationally renowned academic journal "American Chemical Society Journal" published the research results of Professor Liang Gaolin's research group of the School of Chemistry and Materials Science of the University of Science and Technology of China. The article titled "Cell Environment-Differentiated Self-Assembly of Nanofibers". This article reports on a novel "smart" small molecule hydrogel precursor that allows for the differentiation of cells inside and outside the environment and assembly into nanofibers of different structures (J. Am. Chem. Soc., 2016, 138(35), 11128-11131).
The interior of a tumor cell is usually a reducing environment in which glutathione (GSH) is present in the order of millimolar. There are a large number of phosphatase secreted by cells outside the tumor cells. Using this difference, Liang Gaolin's team designed a biocompatible multi-functional small molecule hydrogel precursor. The small molecule's phosphate is cleaved by the extracellular phosphatase, triggering the first stage of self-assembly to form linear nanofibers (see figure below). When these small molecules enter the interior of the cell, the disulfide bond on the small molecule is reduced by GSH due to the presence of GSH, thereby exposing the mercaptothiol structure on the cysteine. The naked aminothiol structure can be efficiently click-condensed with cyanobenzothiazole in a small molecular structure to form a cyclic dimer. Due to the π-π stacking effect, the dimer molecules attract each other, triggering a second-stage self-assembly to form a ring-shaped nanofiber (see the figure below). The realization of the "one stone two birds" design of the small molecule hydrogel precursor is another successful application of the new click condensation reaction (reaction between cysteine ​​and cyanobenzothiazole) discovered and promoted by Professor Liang Gaolin.
